RESUMO

Effects of different ionic strengths (0.2, 0.4, and 0.6 mol/L) and different hydrodynamic cavitation (HC) treatment times (0, 1, 2, 3, and 4 min) on the conformation and gel properties of tilapia myofibrillar proteins (TMP) were investigated. The results showed that the solubility of TMP was significantly enhanced (p < 0.05) with the increase in NaCl concentration, and the gel characteristics were significantly improved. After HC treatment of TMP, the average particle size was significantly reduced (p < 0.05) and solubility was significantly enhanced (p < 0.05) with the increase in treatment time, the internal hydrophobic groups and reactive sulfhydryl groups were exposed. The intrinsic fluorescence spectra showed the unfolding of the spatial tertiary structure of proteins, and the circular dichroism spectroscopy showed the significant reduction in the content of α-helix in the secondary structure of the proteins (p < 0.05). In addition, the WHC and gel strength of the TMP heat-induced gels were enhanced, which improved the microstructure of the gels, and scanning electron microscopy showed that the gel network of the TMP gels became denser and more homogeneous. Dynamic rheology results showed that HC treatment resulted in a significant increase in the final G' and G" values of TMP. In conclusion, HC treatment was able to improve the physicochemical structure and gel properties of TMP at different ionic strengths. This study presents a novel processing technique for the quality maintenance aspect of salt-reduced surimi gel products.

Degenerative bone disorders have a significant impact on global health, and regeneration of articular cartilage remains a challenge. Existing cell therapies using mesenchymal stromal cells (MSCs) have shown limited efficacy, highlighting the necessity for alternative stem cell sources. Here, we have identified and characterized MSX1+ mesenchymal progenitor cells in the developing limb bud with remarkable osteochondral-regenerative and microenvironment-adaptive capabilities. Single-cell sequencing further revealed the presence of two major cell compositions within the MSX1+ cells, where a distinct PDGFRAlow subset retained the strongest osteochondral competency and could efficiently regenerate articular cartilage in vivo. Furthermore, a strategy was developed to generate MSX1+PDGFRAlow limb mesenchyme-like (LML) cells from human pluripotent stem cells that closely resembled their mouse counterparts, which were bipotential in vitro and could directly regenerate damaged cartilage in a mouse injury model. Together, our results indicated that MSX1+PDGFRAlow LML cells might be a prominent stem cell source for human cartilage regeneration.

Polychlorinated dibenzo-p-dioxins (PCDDs), comprising 75 congeners, have gained considerable attention from the general public and the scientific community owing to their high toxic potential. The base-catalyzed hydrolysis of PCDDs is crucial for the assessment of their environmental persistence. Nonetheless, owing to the substantial number of congeners and low hydrolysis rates of PCDDs, conducting hydrolysis experiments proves to be exceedingly time-consuming and financially burdensome. Herein, density functional theory and transition state theory were employed to predict the base-catalyzed hydrolysis of PCDDs in aquatic environments. Findings reveal that PCDDs undergo base-catalyzed hydrolysis in aquatic environments with two competing pathways: prevailing dioxin ring-opening and reduced reactivity in the hydrolytic dechlorination pathway. The resultant minor products include hydroxylated PCDDs, which exhibit thermodynamic stability surpassing that of the principal product, chlorinated hydroxydiphenyl ethers. The half-lives (ranging from 17.10 to 1.33 × 1010 h at pH = 8) associated with the base-catalyzed hydrolysis of PCDDs dissolved in water were shorter compared to those within the water-sediment environmental system. This observation implies that hydroxide ions can protect aquatic environments from PCDD contamination. Notably, this study represents the first attempt to predict the base-catalyzed hydrolysis of PCDDs by using quantum chemical methods.

BACKGROUND: The efficacy of pharmacological and nutritional interventions in individuals at clinical high risk for psychosis (CHR-P) remains elusive. This study aims to investigate the efficacy of pharmacological and nutritional interventions in CHR-P and whether these interventions can enhance the efficacy of psychological treatments. METHODS: We systematically reviewed data from 5 databases until July 24, 2021: PubMed, Web of Science, EMBASE, China National Knowledge Infrastructure, and WanFang Data. The primary outcome was the transition to psychosis. Network meta-analyses were conducted at 3 time points (6, 12, and ≥24 months) considering both pharmacological/nutritional interventions alone and its combination with psychotherapy. RESULTS: Out of 11 417 identified references, 21 studies were included, comprising 1983 participants. CHR-P participants receiving omega-3 polyunsaturated fatty acids treatment were associated with a lower probability of transition compared with placebo/control at 6 months (odds ratio [OR] = 0.07, 95% confidence interval [CI] = .01 to .054), 12 months (OR = 0.14, 95% CI = .03 to .66), and ≥24 months (OR = 0.16, 95% CI = .05 to .54). Moreover, risperidone plus psychotherapy was associated with a lower likelihood of transition at 6 months compared with placebo/control plus psychotherapy, but this result was not sustained over longer durations. CONCLUSION: Omega-3 polyunsaturated fatty acids helped in preventing transitions to psychosis compared with controls. PROSPERO REGISTRATION NUMBER: CRD42021256209.

Equipped with advanced sensors and capable of relaying safety messages to drivers, connected vehicles (CVs) hold the potential to reduce crashes. The goal of this study is to assess the impacts of CV technologies on driving behaviors and safety outcomes in highway crash scenarios under diverse weather conditions, including clear and foggy weather. A driving simulator experiment was conducted and the multigroup structural equation modeling (SEM) was employed to explore the complex interrelationships between the propensity of traffic conflicts, utilization of CV alerts, weather, psychological factors, driving behaviors, and other relevant variables for two different crash locations, namely a straight section and a horizontal curve. Two latent psychological factors including aggressiveness and unawareness were constructed from driving behavior as vehicles passed by crash scenes such as brake, throttle, steering angle, lane offset, and yaw. The SEM can measure latent psychological factors and model interrelationships concurrently through a single statistical estimation procedure. Results of the multigroup SEM showed that CV alerts could significantly reduce the unawareness on a horizontal curve and thus lower the propensity of traffic conflicts. Additionally, the overall effect of foggy weather on conflicts was found to be positive on a horizontal curve, despite the potential benefit of improving situational awareness. In contrast, the single group SEM failed to reveal any significant interrelationships in its structural model by pooling data from both crash locations. The obtained insights can guide the development of driving assistance systems, highlighting the necessity of customization considering weather conditions and location-specific factors.

Copy-number variations (CNVs), which refer to deletions and duplications of chromosomal segments, represent a significant source of variation among individuals, contributing to human evolution and being implicated in various diseases ranging from mental illness and developmental disorders to cancer. Despite the development of several methods for detecting copy number variations based on next-generation sequencing (NGS) data, achieving robust detection performance for CNVs with arbitrary coverage and amplitude remains challenging due to the inherent complexity of sequencing samples. In this paper, we propose an alternative method called OTSUCNV for CNV detection on whole genome sequencing (WGS) data. This method utilizes a newly designed adaptive sequence segmentation algorithm and an OTSU-based CNV prediction algorithm, which does not rely on any distribution assumptions or involve complex outlier factor calculations. As a result, the effective detection of CNVs is achieved with lower computational complexity. The experimental results indicate that the proposed method demonstrates outstanding performance, and hence it may be used as an effective tool for CNV detection.

Nitrogen fixation using electrochemical methods on the surface of single-atom catalysts (SACs) provides a highly feasible strategy for green and low-energy-consumption ammonia (NH3) production. Herein, using density functional theory (DFT) calculations, we explored in detail the potential of monolayer BC3N2 SACs supported with 3d transition metal (TM) atoms (TM@BC3N2) to facilitate nitrogen reduction. The results revealed that the TM@BC3N2 systems exhibited remarkable catalytic activity in the nitrogen-reduction reaction (NRR). The fine NRR activity was related to the just-right bonding/antibonding orbital interactions between the 2π* of N2 and the d orbitals of the TM ions. The nitrogen-adsorption configurations were found to have different activation mechanisms. In addition, the effects of convectively formed convex nitrogen defects (VN) on the interaction between N2 and VN-TM@BC3N2 and the NRR process of VN-TM@BC3N2 were studied, and it was found that VN could fine-tune the reaction efficiency of the eNRR because after N atom dissociation to form VN, the interaction of TM-C3 was enhanced, and the activation of nitrogen and adsorption of NH3 by the TM-active centers were weakened. The present study can be used as a motivation for further experimental and theoretical research of 2D monolayers as NRR electrocatalysts.

OBJECTIVE: This study examined the impact of monitoring instructions when using an automated driving system (ADS) and road obstructions on post take-over performance in near-miss scenarios. BACKGROUND: Past research indicates partial ADS reduces the driver's situation awareness and degrades post take-over performance. Connected vehicle technology may alert drivers to impending hazards in time to safely avoid near-miss events. METHOD: Forty-eight licensed drivers using ADS were randomly assigned to either the active driving or passive driving condition. Participants navigated eight scenarios with or without a visual obstruction in a distributed driving simulator. The experimenter drove the other simulated vehicle to manually cause near-miss events. Participants' mean longitudinal velocity, standard deviation of longitudinal velocity, and mean longitudinal acceleration were measured. RESULTS: Participants in passive ADS group showed greater, and more variable, deceleration rates than those in the active ADS group. Despite a reliable audiovisual warning, participants failed to slow down in the red-light running scenario when the conflict vehicle was occluded. Participant's trust in the automated driving system did not vary between the beginning and end of the experiment. CONCLUSION: Drivers interacting with ADS in a passive manner may continue to show increased and more variable deceleration rates in near-miss scenarios even with reliable connected vehicle technology. Future research may focus on interactive effects of automated and connected driving technologies on drivers' ability to anticipate and safely navigate near-miss scenarios. APPLICATION: Designers of automated and connected vehicle technologies may consider different timing and types of cues to inform the drivers of imminent hazard in high-risk scenarios for near-miss events.

We successfully fabricated two-dimensional metallic CoBi nanoislands on SrTiO3(001) substrate by molecular beam epitaxy, and systematically investigated their electronic structures by scanning tunneling microscopy and spectroscopy in situ at 4.2 K. Coulomb blockade and Coulomb staircases with discrete and well-separated levels are observed for the individual nanoisland, which is attributed to single-electron tunneling via two tunnel junction barriers. They are in excellent agreement with the simulations based on orthodox theory. Furthermore, we demonstrated that the Coulomb blockade becomes weaker with increasing temperature and almost disappears at ~22 K in our variable temperature experiment, and its full-width at half-maximum of dI/dV peaks with temperature is ~ 6 mV. Our results provide a new platform for designing single-electron transistors that have potential applications in future microelectronics. .

There have been some studies conducted about the single factor viscoelasticity of polymer solution or wettability effect on the micro-flow mechanism of polymer flooding. In this paper, the flow mechanism of polymer solution in dual heterogeneous reservoir considering the wettability and gravity was studied. The influences of wettability and rock particle shape on flow characteristics were studied based on the characteristics of saturation and pressure distribution. Compared with the simulation results of polymer flooding in three different rock particle shapes porous media, the oil displacement efficiency of the circular particle model is the highest at 91.57%, which is 3.34% and 11.48% higher than that in the hexagonal and diamond models, respectively. The influence of wettability was studied by the circular particle model. The oil displacement efficiency under water-wet conditions was higher than that under oil-wet conditions. The displacement process considering gravity was affected by the crossflow caused by gravity and viscous force, and the micro-oil displacement efficiency was 9.87% lower than that of non-gravity. Considering the wettability, vertical crossflow will be formed. The oil displacement efficiency under water-wet conditions was 3.9% higher than in oil-wet conditions. The research results can not only expand and enrich the micro-flow mechanism of viscoelastic polymer solution, but also provide reference and guidance for polymer flooding scheme design.

Despite many research efforts on ride-hailing services and taxis, limited studies have compared the safety performance of the two modes. A major challenge is the need for reliable mode-specific exposure data to model their safety outcomes. Moreover, crash frequencies of the two modes by injury severities tend to be spatially and inherently correlated. To fully address these issues, this study proposes a novel multivariate conditional autoregressive model considering measurement errors in mode-specific exposures (MVCARME). More specially, a classical measurement error structure accommodates the uncertainty of estimated mode-specific exposures, and a multivariate spatial specification is adopted to capture potential spatial and inherent correlations. The model estimation is accelerated by an integrated nest Laplace approximation method. The census tracts in the city of Chicago are set as the spatial analysis unit. The mode-specific exposures (vehicle-mile-traveled) in each census tract are estimated by trip assignments using ride-hailing and taxi trip data in 2019. The modeling results indicate that both ride-hailing crashes and taxi crashes are positively associated with transportation factors (e.g., vehicle-mile-traveled, mode-specific vehicle-mile-traveled, and traffic signal numbers), land use factors (i.e., number of educational and alcohol-related sites), and demographic factors (e.g., median household income, transit ratio, and walk ratio). By comparison, the proposed model outperforms the others (i.e., negative binomial models and multivariate conditional autoregressive model) by yielding the lowest deviance information criterion (DIC), Watanabe-Akaike information criterion (WAIC), mean absolute error (MAE), and root-mean-square error (RMSE). According to the results of t-tests, ride-hailing services are found to be prone to a higher risk of minor injury crashes compared with taxis, despite no significant difference between the risks of severe injury crashes. Methodologically, this study adds a robust safety evaluation approach for comparing crash risks of different modes to the literature. At the same time, practically, it provides researchers, practitioners, and policy-makers insights into the safety management of various mobility alternatives.

Salinity stress is a great threat to the growth and productivity of crops, and development of salt-tolerant crops is of great necessity to ensure food security. Although a few genes with natural variations that confer salt tolerance at germination and seedling stage in rice have been cloned, effective intragenic markers for these genes are awaited to be developed, which hinder the use of these genes in genetic improvement of salt tolerance in rice. In this study, we first performed haplotype analysis of five rice salt-tolerant-related genes using 38 rice accessions with reference genome and 4,726 rice germplasm accessions with imputed genotypes and classified main haplotype groups and haplotypes. Subsequently, we identified unique variations for elite haplotypes reported in previous studies and developed 11 effective intragenic makers. Finally, we conducted genotyping of 533 of the 4,726 rice accessions from worldwide and 70 approved temperate geng/japonica cultivars in China using the developed markers. These results could provide effective donors and markers of salt-tolerant-related genes and thus could be of great use in genetic improvement of salt tolerance in rice.

A detailed morphological description and comparative study were conducted on numerous large-sized hamster remains collected from the late Middle Pleistocene Locality 2 of Shanyangzhai (Syz 2), Hebei Province, China. The comparisons reveal that these fossils are highly similar to the extant Tscherskia triton in size and morphology, including the small degree of alternating between the main opposite cusps on M1-3, the presence of axioloph on M3, and mesolophids on m1-2 that are present but seldom reach the lingual margin of the teeth, among other features. However, minor differences between the two still exist. Consequently, all these fossils are designated as a chronosubspecies of the extant species, T. triton varians comb. nov. The skull and molar morphologies of Cricetinus varians and T. triton were meticulously compared to resolve the long-standing debate regarding the validity of Cricetinus Zdansky, 1928 and C. varians Zdansky, 1928. The findings indicate that the differences between the two are slight; as a result, C. varians can only be considered a chronosubspecies of T. triton, i.e., T. triton varians comb. nov., and Cricetinus should be recognized as a junior synonym of Tscherskia. We tentatively propose that, among the seven species once referred to Cricetinus, C. europaeus, C. gritzai, C. janossyi, and C. koufosi should be reassigned to Tscherskia, while C. beremendensis should be transferred to Allocricetus, and C. mesolophidos to Neocricetodon. Excluding Tscherskia sp. from the Late Pliocene Youhe fauna, there are no reliable Tscherskia fossils in China earlier than the Middle Pleistocene. Based on the current evidence, Tscherskia may have originated from Neocricetodon during the Early Pliocene in Europe and subsequently spread to Asia. T. triton is its sole surviving representative, which now exclusively inhabits East Asia.

DNA is highly vulnerable to spontaneous and environmental timely damage in living cells. DNA damage may cause genetic instability and increase cancer risk if the damages are not repaired timely and efficiently. Human cells possess several DNA damage response (DDR) mechanisms to protect the integrity of their genome. Clarification of the mechanisms underlying the DNA damage response following lethal damage will facilitate the identification of therapeutic targets for cancers. Histone post-translational modifications (PTMs) have been indicated to play different roles in the repair of DNA damage. In this context, histone PTMs regulate recruitment of downstream effectors, and facilitate appropriate repair response. This review outlines the current understanding of different histone PTMs in response to DNA damage repair, besides, enumerates the role of new type PTMs such as histone succinylation and crotonylation in regulating DNA damage repair processes.

Single nucleotide variants (SNVs) are very common in human genome and pose a significant effect on cellular proliferation and tumorigenesis in various cancers. Somatic variant and germline variant are the two forms of SNVs. They are the major drivers of inherited diseases and acquired tumors respectively. A reasonable analysis of the next generation sequencing data profiles from cancer genomes could provide crucial information for cancer diagnosis and treatment. Accurate detection of SNVs and distinguishing the two forms are still considered challenging tasks in cancer analysis. Herein, we propose a new approach, LDSSNV, to detect somatic SNVs without matched normal samples. LDSSNV predicts SNVs by training the XGboost classifier on a concise combination of features and distinguishes the two forms based on linkage disequilibrium which is a trait between germline mutations. LDSSNV provides two modes to distinguish the somatic variants from germline variants, the single-mode and multiple-mode by respectively using a single tumor sample and multiple tumor samples. The performance of the proposed method is assessed on both simulation data and real sequencing datasets. The analysis shows that the LDSSNV method outperforms competing methods and can become a robust and reliable tool for analyzing tumor genome variation.

The adsorption of five small gas molecules (CO, CO2, NO, NO2, and NH3) on transition metal (TM)-modified ZrSe2 monolayers (Au-ZrSe2 and Pt-ZrSe2) are studied based on first principles. The adsorption structure, adsorption energy (Eads), electron transfer (Qt), and density of states (DOS) with intrinsic ZrSe2, Au-ZrSe2, and Pt-ZrSe2 monolayers are discussed, and their sensing performance is evaluated. The results show that the electrical conductivity of ZrSe2 is obviously increased after Au and Pt atom modification. The intrinsic ZrSe2 adsorbs the five kinds of gas molecules weakly, while ZrSe2 modified by the Au or Pt atom improves the adsorption of the gas molecules in different degrees. Au-ZrSe2 has the best adsorption effect on NO2 gas molecules, while Pt-ZrSe2 shows strong sensitivity to CO gas molecules. Moreover, Au-ZrSe2 and Pt-ZrSe2 are of great significance for the adsorption sensing mechanism and also offer prospective materials for the advancement of gas-sensitive sensors.

Since the oxygen reduction reaction (ORR) is the cathodic reaction of energy storage and conversion devices such as fuel cells and metal-air batteries, the search for catalysts with high-performance toward ORR has become the focus of attention. Transition metal dichalcogenides (TMDs) have the advantages of easy integration, inexpensive, harmless, good stability, and have vast application prospects in stabilizing the single atom. Hence, in this study, we investigated the feasibility of several 4d/5d single transition metals (TM = Rh, Pd, Ag, Ir, Pt, and Au) doped with CrSe2 for ORR electrocatalysis on the basis of density functional theory (DFT) calculations. Our results demonstrated that most of the TM-doped systems are stable, exhibiting metal conductivity, and can well activate the adsorbed O2. Interestingly, compared with end-on adsorption configuration, O2 is more likely to be adsorbed on the catalysts by a side-on adsorption configuration. Among all the candidate ORR catalysts, Pd-doped and Pt-doped CrSe2 have the best catalytic performance with a low overpotential of 0.43 and 0.50 V, respectively, making them good ORR candidate high performance catalysts. Our DFT work helps to understand the interaction between the O2 molecule and transition metal single atoms on CrSe2 supports and provides ideas for designing stable and efficient ORR catalysts.

Male infertility is a global issue that seriously affects reproductive health. This study aimed to understand the underlying causes of idiopathic non-obstructive azoospermia (iNOA), which is a type of male infertility with unknown origins that accounts for 10-15% of cases. By using single-cell analysis techniques, we aimed to uncover the mechanisms of iNOA and gain insight into the cellular and molecular changes in the testicular environment. In this study, we performed bioinformatics analysis using scRNA-seq and microarray data obtained from the GEO database. The analysis included techniques such as pseudotime analysis, cell-cell communication, and hdWGCNA. Our study showed a significant difference between the iNOA and the normal groups, indicating a disorder in the spermatogenic microenvironment in iNOA. We observed a reduction in the proportion of Sertoli cells and blocked germ cell differentiation. Additionally, we found evidence of testicular inflammation related to macrophages and identified ODF2 and CABYR as potential biomarkers for iNOA.

Lane changes are complex driving behaviors and frequently involve safety-critical situations. This study aims to develop a lane-change-related evasive behavior model, which can facilitate the development of safety-aware traffic simulations and predictive collision avoidance systems. Large-scale connected vehicle data from the Safety Pilot Model Deployment (SPMD) program were used for this study. A new surrogate safety measure, two-dimensional time-to-collision (2D-TTC), was proposed to identify the safety-critical situations during lane changes. The validity of 2D-TTC was confirmed by showing a high correlation between the detected conflict risks and the archived crashes. A deep deterministic policy gradient (DDPG) algorithm, which could learn the sequential decision-making process over continuous action spaces, was used to model the evasive behaviors in the identified safety-critical situations. The results showed the superiority of the proposed model in replicating both the longitudinal and lateral evasive behaviors.
